- MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000. Protein phosphatase type 2A (PP2A) is a major eukaryotic serine/threonine protein phosphatase (PPase). PP2A is highly conserved and often exist as a heterotrimeric enzyme composed of a catalytic (C) subunit, a structural (A) subunit, and a variable regulatory/targeting (B-type) subunit1. Recent studies show that PP2A plays important roles in cell cycle regulation, signal transduction, cell differentiation, and transformation1-3. Its activity is also related to several diseases, including neurodegenerative diseases and cancer. The enzymatic activity and intracellular localization of PP2A are determined by three distinct families of cellular regulatory subunits, B, B’, and B’’. The B’ subunit, also known as B56, is most diverse, consisting of five isoforms (alpha, beta, gamma, delta, and epsilon).4,5 Based on a conserved epitope, shared with the B’ subunits, striatin (PR110) and S/G2 nuclear autoantigen (SG2NA; PR93) were identified as new members of a potential B’’’ subunit family. A recent study demonstrated that PP2A B56 gamma subunits binds both APC and cyclin G, and that suppression of this PP2A B subunit may directly perturb specific signaling pathways. Alterations in PP2A B56 gamma subunit function may contribute to cancer development in spontaneously arising human tumors.6 Applications: Suitable for use in Western Blot. Other applications have not been tested. Recommended Dilution: Western Blot: 1-3ug/ml. Detects a band of ~60kD in human PP2A B56γ-transfected 293T cell lysates. Optimal working dilutions to be determined by researcher. Positive Control: PP2A B56gamma-transfected 293T cell lysates. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
