- Extra Details:
- Proteinase 3 (PR3), a constituent of azurophil granules of neutrophils (polymorphonuclear cells, PMNs), is the target antigen for most anti-neutrophil cytoplasmic antibodies (c-ANCA) in Wegener’s Granulomatosis (WG). Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilutions: ELISA: 1:100 Optimal dilutions to be determined by the researcher. Autoantibody Activity: Anti-Proteinase 3 Activity by ELISA: Current lot value: 85EU/ml MPO: 0EU/ml Interpretation of Results: Negative: <0.8EU/ml, Equivocal: 0.8-1.19EU/ml, Positive: >1.2 EU/ml Other Autoantibody Activity in the Serum: The serum exhibits no reactivity at a dilution of 1:100 when tested against purified SSA, SSB, SCL, Sm, Sm/RNP, Jo-1, mitochondrial or ribosomal antigens in an ELISA system designed to detect IgG antibody subclasses. Antigen Result (EU/ml) SSA 2 SSB 0 SCL 0 Sm 0 Sm/RNP 3 Jo-1 6 Interpretation of Results: Negative: <16EU/ml, Equivocal: 16-20EU/ml, Positive: >20EU/ml Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
