- Alkaline phosphatase, liver/bone/kidney encoded by the ALPL gene, also known as tissue nonspecific alkaline phosphatase (TNAP), is related to, but distinct from, intestinal/ALPI, placental/ALPP and placental-like/ALPPL2 alkaline phosphatases. ALPL is highly expressed in undifferentiated human embryonic stem cells, embryonic carcinoma cells, and embryonic germ cells. Mutations in this enzyme have been linked directly to hypophosphatasia, a disorder that is characterized by hypercalcemia and skeletal defects. The amino acid sequence of human ALPL is 90%, 89% and 75% identical to that of rat/mouse, canine and chicken. Applications: Suitable for use in Western Blot and ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 1-2ug/ml. The detection limit is ~10 ng/lane under non-reducing and reducing conditions. Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~2ng/well. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
