- Extra Details:
- PERK, a type 1 ER membrane kinase, mediates eIF2-alpha phosphorylation at Ser51 during the UPR (unfolded protein response). Protein synthesis is inhibited, thereby reducing the burden of protein substrate for the ER folding and degradation mechanism. Phosphorylation of eIF2 alpha also selectively promotes the expression of UPR target genes such as Chop and BiP. PERK may also play a role in tumor cell adaptation to hypoxic stress by regulating the translation of angiogenic factors necessary the development of functional microvessels. Mutations in PERK are responsible for the rare autosomal-recessive disorder, WRS (Wolcott-Rallison syndrome). Applications: Suitable for use in Western Blot and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
