- Extra Details:
- PERK, an eIF2 alpha kinase, is a transmembrane protein resident in the endoplasmic reticulum (ER) membrane and couples ER stress signals to translation inhibition. ER stress increases the activity of PERK, which then phosphorylates eIF2 alpha, resulting in reduced translation. PERK-deficient mice have defects in pancreatic beta cells several weeks after birth, suggesting a role for PERK-mediated translational control in protecting secretory cells from ER stress. PERK activation during ER stress correlates with autophosphorylation of its cytoplasmic kinase domain. Phosphorylation of PERK at Thr980 can serve as a marker for its activation status. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000. Incubate membranes with diluted antibody, 1X TBS, 5% BSA, 0.1% Tween-20 at 4ºC with gentle shaking overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
