Extra Details:
Perforin, originally cloned from mouse cytotoxic T-lymphocyte (CTL), is a 555-amino acid protein with a 21-amino acid signal peptide and is a potent cytolytic pore-forming protein. It is a specific marker of functionally active cytotoxic T-lymphocytes (CTLs) and natural killer (NK) cells. Fas ligand and perforin are the two key effector mechanisms in T cell-mediated cytotoxicity. These molecules mediate cytolysis of target cells by membrane damage and apoptosis, an important mechanism in the immune defense against tumors and virus infection. Perforin expression is constitutive on NK cells, but increases in resting XD8+ cytotoxic cells upon activation. Applications: Suitable for use in Immunohistochemistry. Other applications have not been tested. Recommended Dilution: Immunohistochemistry (Formalin/paraffin): 1:10 for 60 min at RT . Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA, pH 8, for 10-20 min followed by cooling at RT for 20 min. Optimal dilution to be determined by the researcher. Positive Control: Lymphoma Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
