- Pepsinogen I, a zymogen, or proenzyme secreted by Chief cells in the stomach, is cleaved to form pepsin both in an autocatalytic fashion and by pepsin itself. In humans there are two related forms of pepsin, Pepsinogen I, also known as Pepsinogen A, and Pepsinogen II, also known as Pepsinogen B or progastricsin. A protein precursor to pepsin, Pepsinogen I or Pepsinogen A is secreted exclusively by the chief cells of the gastric corpus. The levels of Pepsinogen I correlate directly with the active number of chief cells in this area. In cases of severe atrophic gastritis of the corpus region, the chief cells will be destroyed and the levels of Pepsinogen I will fall dramatically. Applications: Suitable for use in ELISA, Immunohistochemistry and RIA. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry: Paraffin sections. Antigen retrieval using heat treatment prior to stainin of paraffin sections. Sodium citrate pH 6.0 is recommended. Optimal dilutions to be determined by the researcher. Affinity Constant: 4x10e10L/mol Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Note: Applications are based on unconjugated antibody.
