Extra Details:
MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000. Pigment epithelium-derived factor (PEDF) was first identified as a 50kD secreted protein in conditioned medium from cultured fetal human retinal pigment epithelium (RPE) cells. More recently, PEDF was shown to have neurotrophic activity in the retina in vivo. PEDF is also a neurotrophic and neuroprotective factor in other systems, including cultured cerebellar granule cells, primary hippocampal neurons, spinal cord motor neurons and amphibian photoreceptors cultured in the absence of RPE cells. The binding of PEDF to retinoblastoma and cerebellar granule cells may be receptor-mediated. PEDF has been shown to be a potent inhibitor of angiogenesis in endothelial cell cultures and in rat cornea and retina in vivo. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: detects PEDF in 40ug of RIPA lysates from Y79 cells. 40ug of Y79 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with P3140. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb I1904-46Q: IgG, H&L (HRP) Pab Gt x Rb Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.  Note: Applications are based on unconjugated antibody.
