- Applications: Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry, Blocking of receptor-ligand interaction and Direct ELISA. Other applications not tested. Recommended Dilution: Western Blot: 0.1-0.2ug/mL with the appropriate secondary reagents to detect mouse PD-L2. The detection limit for rmPD-L2 is ~2ng/lane under non-reducing and reducing conditions. Immunohistochemistry: 2-15ug/ml will detect PD-L2 in cells and tissues. Flow Cytometry: on Tim-1 stimulated RAW 264.7 cells for use in flow cytometry. Dilute this antibody to 50ug/mL and add 10ul of the diluted solution to 1-2.5 x 10e5 cells in a total reaction volume not exceeding 200ul. The binding of unlabeled antibodies may be visualized by adding 10ul of a 25ug/mL stock solution of a secondary developing reagent such as anti-goat IgG conjugated to a fluorochrome. Blocking of receptor-ligand interaction: ~1-4ug/mL of this antibody will block 50% of the binding of 1ug/mL of rmPD-1 to immobilized rmPD-L2/Fc chimera (100ul of a 1ug/mL solution was coated in each well) in a functional ELISA. Direct ELISA: 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse PD-L2. The detection limit for rmPD-L2 is ~1.2 ng/well. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
