Extra Details:
Cell-mediated immune responses are initiated by T lymphocytes that are themselves stimulated by cognate peptides bound to MHC molecules on antig en-presenting cells (APC). T-cell activation is generally self-limited as activated T cells express receptors such as PD-1 (also known as PDCD-1) that mediate inhibitory signals from the APC. PD-1 can bind two different but related ligands, PDL-1 and PDL-2. Upon binding to either of these ligands, signals generated by PD-1 inhibit the activation of the immune response in the absence of "danger signals" such as LPS or other molecules associated with bacteria or other pathogens. Evidence for this is seen in PD1-null mice who exhibit hyperactivated immune systems and autoimmune diseases. Applications: Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry and Immunofluorescence. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml. See P3120-60A for control peptide. Immunofluorescence: 10ug/ml Immunohistochemistry (paraffin): 5ug/ml. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20min. followed by cooling at RT for 10-20min. Immunocytochemistry: 10ug/ml Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1903-30U: IgG, H&L, X-adsorbed (HRP) ShxRb Recommended Positive Control Tissue: Human brain Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
