MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. CD279 is a single-pass type 1 membrane protein belonging to the CD28 family, and functions mainly as a negative regulator of T-cell activation. CD279 has two specific ligands; CD274 (PD-L1) and CD273 (PD-L2), and their interaction is key in the balance between stimulatory and inhibitory signals needed for effective immune responses to microbes and self-tolerance. CD279 is inducibly expressed by T-cells, B-cells, NK-T-cells and monocytes upon activation. Loss of CD279 function has been associated with a number of autoimmune diseases, including rheumatoid arthritis, type I diabetes and ankylosing spondylitis. Recent studies suggest that CD279 could be targeted therapeutically in the treatment of HIV infection to reduce T-cell exhaustion. Applications: Suitable for use in FLISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: Detects a band at ~45kD in THP-1 cell lysates. Immunohistochemistry (Paraffin): Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20min. followed by cooling at RT for 10-20min. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1903-30U: IgG, H&L, X-adsorbed (HRP) ShxRb Recommended Positive Control Tissue: Human brain Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
