Extra Details:
CD279 is a single-pass type 1 membrane protein belonging to the CD28 family, and functions mainly as a negative regulator of T-cell activation. CD279 has two specific ligands; CD274 (PD-L1) and CD273 (PD-L2), and their interaction is key in the balance between stimulatory and inhibitory signals needed for effective immune responses to microbes and self-tolerance. CD279 is inducibly expressed by T-cells, B-cells, NK-T-cells and monocytes upon activation. Loss of CD279 function has been associated with a number of autoimmune diseases, including rheumatoid arthritis, type I diabetes and ankylosing spondylitis. Recent studies suggest that CD279 could be targeted therapeutically in the treatment of HIV infection to reduce T-cell exhaustion. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: Detects a band at ~45kD in THP-1 cell lysates. Immunohistochemistry (Paraffin): Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20min. followed by cooling at RT for 10-20min. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1903-30U: IgG, H&L, X-adsorbed (HRP) ShxRb Recommended Positive Control Tissue: Human brain Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
