Extra Details:
Proliferating cell nuclear antigen (PCNA) is a 36kD nuclear protein associated with the cell cycle, a nuclear protein vital for cellular DNA synthesis. Proliferating cell nuclear antigen was originally identified by immunofluorescence as a nuclear protein whose appearance correlated with the proliferate state of the cell. PCNA is required for replication of DNA in vitro and has been identified as the auxiliary protein (cofactor) for DNA polymerase delta. The anti-PCNA antibodies react with the nuclei of proliferating cells. PCNA is essential for cellular DNA synthesis and is also required for the in vitro replication of simian virus 40 (SV40) DNA where it acts to coordinate leading and lagging strand synthesis at the replication fork. The PCNA protein may fulfil several separate roles in the cell nucleus associated with changes in its antigenic structure. Applications: Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.1-1ug/ml Flow Cytometry: 1:100 Immunohistochemistry (formalin fixed paraffin embedded sections only): 5-50ug/ml. May be used on material fixed in a wide range of fixatives including formalin (buffered and unbuffered), methacarn and Bouin's reagent. The time of fixation can markedly affect the intensity of PCNA immunoreactivity. Staining is reduced (and may be abolished) if sections are baked onto glass slides. Air-drying overnight on poly-L-lysine coated slides is recommended. 60 minute incubation at 25°C with standard ABC technique is recommended. Optimal dilutions to be determined by the researcher. Positive Control: Tonsil or reactive lymph node. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
