- Paxillin can be diffusely distributed in the cytoplasm, colocalized with actin at the cell periphery, or aggregated at focal adhesions. Paxillin has several motifs which allow its interaction with other proteins. These motifs include LD motifs, LIM domains, an SH3 domain binding site, and SH2 domain binding sites. Paxillin complexes with proteins of focal adhesions including vinculin, talin, with the kinases FAK, PYK2, Src, and Csk, with ARF GTPase activating proteins, and with the adaptor protein v-Crk. Paxillin is phosphorylated at several residues, including tyrosine 31, tyrosine 118, tyrosine 181. The phosphorylation of paxillin appears to be important for several biological functions including S-phase transition and neurite extension. Applications: Suitable for use in ELISA, Immunohistochemistry. Immunocytochemistry, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:10,000. Optimal dilution determined by the researcher. Control Lysate: A431 Lysate: Prepared from a human epidermoid carcinoma cell line. Cell lysate is available to be used as a positive control in Western blotting. Cell lysate is provided at a concentration of 1ug/ml in SDS-PAGE buffer (62mM Tris pH 6.8, 2% SDS, 0.9% b- mercaptoethanol, 0.003% bromphenol blue and 5% glycerol). 5–10ul/lane is sufficient for Western blot Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
