Extra Details:
Several peptides associated with feeding behavior have been reported recently. Orexins (Orexin A and Orexin B) are a family of hypothalamic neuropeptides selectively expressed in the hypothalamus and are derived from the same precursor (Prepro-orexin) by proteolytic cleavage. Prepro-orexin is 130 amino acid long peptide with a putative 33 AA secretory sequence, a hydrophobic core followed by residues with small polar side chains. The expression was detected in brain and to a small extent in testis. These neuropeptides bind and activate two closely related Orexin receptors—G-protein coupled receptors (GPCRs). Rat and human Orexin 1Receptor (Hypocretin 1 Receptor) are 416 aa, and 425aa, respectively. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: ELISA: 1:10,000-1:100,000 (Use control peptides to confirm antibody specificity) Western Blot: 1-10ug/ml using chemiluminescence Immunohistochemistry: 2-20ug/ml. Antigen retrieval was carried out by pressure cooker unmasking for 2 min in 0.01M sodium citrate buffer (pH 6.0) Optimal dilution determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
