Extra Details:
OSM is a 209aa pleiotropic cytokine, originally isolated from PMA (phorbol 12-myristate 13-acetate) treated U937 leukemia cells and mitogen-activated T lymphocytes. It was identified by the ability to inhibit the replication of various human tumor cell lines, but has also been shown to exert a stimulatory effect on fibroblasts, smooth muscle cells and Kaposi's sarcoma cells, and to induce the release of certain cytokines by endothelial cells. OSM signals through binding with low affinity to the gp130 protein present in two cell surface receptors: type I consisting of gp130 and LIFR (leukocyte inhibitory factor receptor) which also acts as a receptor for LIF, and type II consisting of gp130 and OSMR, which mediates OSM specific activities. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilutions: ELISA (Sandwich): 0.25-1ug/ml using 100ul/well antibody solution. Allows the detection of at least 0.2-0.4ng/well. Western Blot: 0.1-0.2ug/ml. The detection limit is 1.5-3ng/lane, under either reducing or non-reducing conditions. Optimal dilution to be determined by researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS, 0.1% BSA. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
