Extra Details:
Human Notch-2 is a 300kD member of the notch family of transmembrane (TM) proteins. It is synthesized as a 2446 amino acid (aa) type I TM glycoprotein that undergoes Golgi processing to generate a 180kD disulfide-linked extracellular domain (ECD) (likely aa25-1538) and a 110kD membrane bound truncated segment (likely aa1539-2471). Upon binding to Delta1, Jagged1, or Jagged2, the 110kD segment undergoes two cleavages, the second which generates an NICD (notch intracellular domain) (aa1697-2471), a potential nuclear transcription factor. In the nucleus, NICD regulates CSL pathways. When NICD is phosphorylated, it is deactivated. Applications: Suitable for use in Western Blot and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
