- NOD2 (nucleotide-binding oligomerization domain containing 2), also known as the caspase recruitment domain family, member 15 (CARD15), is a member of the NOD-like Receptor (NLR) family of proteins and plays an important role in the immune system. It is an intracellular pattern recognition receptor and is linked to inflammatory diseases such as inflammatory bowel disease/Crohn′s Disease and Blau syndrome. NOD2 has two caspase recruitment domains (CARDs) and eleven leucine-rich repeats (LRRs). The protein is primarily expressed in the peripheral blood leukocytes. It plays a role in the immune response by recognizing the bacterial molecules. Recently it is has been found to interact with NLRC4 and bind to MAVS in response to ssRNA or viral RNA treatment and activate the IFN response. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:100-1:2000. Tested on NOD2 transfected 293T lysates and HT29 human colorectal adenocarcinoma lysates. Endogenous Immunohistochemistry (Paraffin): 1:25 detected NOD2 in hyperplastic colorectal carcinoma tissue. Immunoprecipitation: 1:10-1:500 Optimal dilutions to be determined by the researcher. Control: Hyperplastic colorectal carcinoma tissue Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
