- The ion channels activated by glutamate are divided into two classes. Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR) while those activated by kainate and a-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid (AMPA) are known as kainate/AMPA receptors (K/AMPAR). NMDA receptors are among the most studied receptors in neuroscience because they are involved in neuronal cell development and plasticity, a cellular correlate for learning. NMDA receptors are also implicated in several disorders of the central nervous system including epilepsy and ischemic neuronal cell death. NMDA receptors also appear to be a target for ethanol at physiological concentrations and therefore may play a significant role in alcoholism. Applications: Suitable for use in Immunohistochemistry, Immunocytochemistry and Western Blot. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (Light and EM): Using 4% paraformaldehyde and 0.125% glutaraldehyde fixed tissue. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
