Extra Details:
Mammalian NHE (Na+/H+ Exchanger) is an integral membrane protein that is known to regulate intracellular pH by removing a proton in exchange for an extracellular sodium ion. There are nine known isofoms of NHE. The best characterized is NHE1, involved not only in intracellular pH control, but cell-volume control, cytoskeletal organization, heart disease and cancer. Transport by the NHE plays a pivotal role in the damage caused to the human myocardium during and following a myocardial infarction, and it is considered to represent a key step in the oncogenic transformation of cancerous cells. NHE comprises an N-terminal membrane domain that transports ions, and a C-terminal cytoplasmic domain that regulates activity and mediates cytoskeletal interactions. NHE2 - NHE5 isoforms are localized to the plasma membrane but exhibit greater tissue- specificity. NH2 and NH3 are more highly expressed in the kidney and intestine. Structurally, a single transmembrane segment from NHE1 has been solved, and the structure of bacterial Na+/H+ antiporter NhaA has been elucidated. Applications: Suitable for use in Western Blot. Works poorly in Immunohistochemistry and Immunocytochemistry. Not suitable for use in Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 1:500 detected NHE1 on 10ug of human kidney lysates. Human kidney lysate was resolved by electrophoresis, transferred to PVDF membrane and probed with N2015-02 (1:500). Proteins were visualized using an IgG (HRP) Gt xMo* and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. *Recommended Secondary Antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.  Note: Applications are based on unconjugated antibody.
