Extra Details:: In normal tissues and in a variety of myeloproliferative disorders, myeloid cells of both neutrophilic and eosinophilic types, at all stages of maturation, exhibit strong cytoplasmic reactivity for MPO. It is readily detected in myeloblasts and immature myeloid cells of acute myelogenous leukemia, progranulocytic leukemia, monomyelocytic leukemia, erythro-leukemia, myeloblastomas, and other hematopoietic disorders. Erythroid precursors, megakaryocytes, lymphoid cells, mast cells, and plasma cells are nonreactive. Applications: Suitable for use in ELISA and Immunofluorescence. Other applications not tested. Recommended Dilutions: Immunofluorescence: Exhibits a P-ANCA pattern ELISA: 1:100 Optimal dilution determined by the researcher. Autoantibody Activity: Anti-MPO Activity by ELISA: As reported (lot-specific). Current lot value: 96EU/ml Interpretation of Results: <4EU/ml: Negative; 4.1-5.9EU/ml: Equivocal; >6EU/ml: Positive Other Autoantibody Activity in the Serum: The serum exhibits no reactivity at a dilution of 1:100 when tested against purified SSA, SSB, SCL, Sm, Sm/RNP, Jo-1, mitochondrial or ribosomal antigens in an ELISA system designed to detect IgG antibody subclasses. Antigen Result (EU/ml) SSA, SCL, Sm 2 SSB 3 Sm/RNP, Jo-1 1 Interpretation of Results: Negative: <16EU/ml, Equivocal: 16-20EU/ml, Positive: >20EU/ml Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
