- MSH2 is a protein of 934 aa (~100kD) localized to the cell nucleus. MSH2 functions as one of the four major DNA mismatch repair genes along with MLH1, PMS1 and PMS2. Somatic mutations in the human MSH2 gene contribute to the development of sporadic colorectal carcinomas while germline mutations are responsible for approximately 50% of inherited non-polyposis colorectal carcinomas (HNPCC). Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot (chemiluminescence): 0.2ug/ml Immunoprecipitation: 1ug/sample Immunohistochemistry(Frozen): 2ug/ml Immunohistochemistry(Paraffin): 2ug/ml (staining of formalin fixed, paraffin embedded tissue requires Heat induced epitope retrival (HIER) pretreatment) Optimal dilutions to be determined by the researcher. Positive Control: Immunoprecipitation and Western Blot: HCT116 or SW480 cells Immunohistochemistry: Colon tissue. Negative Control: Immunoprecipitation and Western Blot: LoVo cells Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
