Extra Details:
Human Mps1 (hMps1), the human homolog of yeast Mps1p, is a cell cycle-regulated kinase with maximal activity during M phase. It is localized to kinetochores, and its activity and phosphorylation state increase upon activation of the mitotic checkpoint. hMps1 is required for human cells to undergo checkpoint arrest in response to microtubule depolymerization. It is reported that hMps1 is required for the spindle assembly checkpoint but not for centrosome duplication. Recent studies have shown that the overexpression of mMps1p in mice caused reduplication of centrosomes during S phase arrest. Cdk2, which acts as a regulator of centrosome duplication, regulates the stability of Mps1p during S phase. Inhibition of Cdk2 prevents centrosome reduplication, destabilizes mMps1p, and causes its subsequent loss from centrosomes. Studies in yeast models have established that other functions of Mps1 include meiotic spindle pole body duplication, chromosome segregation, and spore wall formation. Applications: Suitable for use in Immunofluorescence, Immunoprecipitation, Western Blot and FLISA. Other applications have not been tested. Recommended Dilution: Immunofluorescence: 5-10ug/ml Immunoprecipitation: 5-10ug/ IP reaction Western Blot: 1-3ug/ml FLISA: 0.1-1ug/ml Note: For immunofluorescent assays, it is recommended that cells are fixed in 4% paraformaldehyde in PBS and permeabilized with 0.2% Triton X-100. Methanol and acetone fixatives can also be used. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
