- Metabotropic glutamate receptors have been classified into three groups. In general, class I receptors (mGluR1 and mGluR5) mediate the excitation of neurons, whereas activation of class II (mGluR2 and mGluR3) and class III (mGluR4a, mGluR7a, mGluR7b and mGluR8) results in a depression of synaptic transmission. Class I mGluRs are localized in the postsynaptic membrane and are positively coupled to phospholipase C (PLC). Class II and class III mGluRs localized to the presynaptic membrane and are negatively coupled to adenylate cyclase. It has been speculated that mGluR2 and/or mGluR3 may play a role in the modulation of the mossy terminal function. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.25-1ug/ml detects a single band at ~140kD. Immunocytochemistry: 0.5-2ug/ml using paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
