- Extra Details:
- Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases. Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K. Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells Applications: Suitable for use in Western Blot. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.5-2ug/ml detects phospho-LAT (Tyr191) in RIPA lysates from Jurkat cells stimulated with C305. Jurkat cells were incubated without or with, a monoclonal antibody that binds Jurkat T-cell receptors. Cell lysates were prepared, resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-LAT (Tyr191) (2ug/ml), or anti- LAT (1ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
