Extra Details:
Laminin subunit Alpha-1 (LAMA1) is a secreted 400kD extracellular matrix glycoprotein that contributes to the formation of basement membrane Laminin isoforms 1 and 3. It is one of three subunits (alpha, beta, and gamma) that interact via their coiled-coil domains to form the approximately 800kD cruciform, disulfide-linked, Laminin heterotrimer. The 3058 amino acid (aa) residue mature human alpha-1 chain contains an N-terminal Laminin VI domain (aa18-269), followed by domains V through III containing 17 EGF-like repeats, the coiled-coil domains II and I, and five globular, Laminin G-like domains. Over aa22-269, human Laminin alpha-1 shares 95% and 91% aa sequence identity with canine and mouse alpha-1 chain, respectively. Applications: Suitable for use in Western Blot, Immunohistochemistry, Flow Cytometry and Direct ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 0.1ug/ml Immunohistochemistry: 5-15ug/ml using immersion fixed frozen sections of mouse embryo. Flow Cytometry: 10ul labels10e6 cells. Validated on U2OS human osteosarcoma. For intracellular staining, cells must first be fixed and permeabilized. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
