- IL-17D, also called IL-27, is one of six IL-17 family members. All are homodimers with conserved cysteines that form a cystine knot structure. IL-17D (Accession # AF479775) is a 52 kD dimer expressed by resting CD4 + T cells and CD19 + B cells that acts on endothelial cells to stimulate the release of IL-6, IL-8, and GM-CSF. With other IL-17 family members, IL-17D recruits neutrophils to sites of inflammation. Applications: Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and ELISA. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml detects recombinant human IL-17D under non-reducing conditions only. Flow Cytometry: 2.5ug/10e6 cells detects human Interleukin 27 in peripheral blood mononuclear cells fixed with paraformaldehyde and permeabilized with saponin Immunohistochemistry: 8-25ug/ml using immersion fixed paraffin-embedded sections of human heart. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
