- IL-21 is a 155 amino acid pro-cytokine, reduced to 133 anino acids in the secreted mature form. IL-21 is a potent immunoregulatory cytokine and member of the IL-15/IL-21 family, expressed and secreted by activated CD4+ T cells. Two potential isoforms are generated by alternative splicing and differing is sequence at the C-terminus (UniProt: Q9HBE4). IL-21 enhances the proliferation/differentiation of stimulated B-cells, the proliferation of bone marrow progenitor cells, and increases the activity of NK cells and disease-specific CD8+ T-cells. IL-21 signals through binding to the specific type I cytokine receptor IL-21R, coupled with the common cytokine receptor g chain (gc), initiating the activation of the JAK/STAT signalling pathway. Applications: Suitable for use in ELISA, Immunocytochemistry and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 1:50,000 Immunocytochemistry: using human tonsil Immunohistochemistry: 1:250 Optimal dilution determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
