- Interleukin 6 Receptor (IL-6 R), is a transmembrane protein that associates with the signal- transducing subunit gp130 to form a high affinity receptor complex for IL-6. It is expressed on T cells, activated B cells, and peripheral monocytes. A soluble form of IL-6 R can be released by proteolytic cleavage. Within the extracellular domain, human IL-6 R shares 52% aa sequence identity with mouse and rat IL-6 R. Applications: Suitable for use in Neutralization, Western Blot and ELISA Other applications have not been tested. Recommended Dilutions: Neutralization of Human IL6 R Bioactivity: ~0.04-0.2ng/ml will neutralize 50% of the bioactivity due to 20ng/ml or recombinant human IL-6R and 20ng/ml of recombinant human IL-6. Recombinat human IL-6R enhances recombinant human IL-6-induced growth inhibition of M1 cells in a dose-dependent manner. The ED50 for this effect is typically 5-15ng/ml in the the presence of 20ng/mk of recombinant human IL-6. Western Blot: 0.1ug/ml under reducing conditions. ELISA Capture: 2-8ug/ml. Use biotinylated human IL-6 R detection antibody and recombinant human IL-6 R as the standard. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
