- Extra Details:
- The cytokine human interferon-gamma (IFN-gamma) is a glycosylated protein that exists as a homodimer of 34kD in its biologically active form. IFN-gamma is expressed upon activation by TH1 cells CD8+ cells and NK cells (2). Secreted IFN-gamma is a potent inhibitor of in vitro hematopoiesis and appears to serve as an important regulator of lymphocyte and macrophage function by exerting antiproliferative effects (1). Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Protocol: 1. Ficoll prepared human peripheral blood lymphocytes were stimulated 4 hours in the presence of 50ng/ml PMA, 2uM ionomicin and 5uM monensin. 2. Cells were then harvested, washed and fixed for 30 min with 2% buffered formaldehyde after which they were washed two times. 3. Fixed cells were permeabolized 10 minutes in a buffer containing 0.3% Saponin. Subsequent reagent incubations and washes were done using this buffer. 4. Fivex10e5 cells per tube were preincubated 5 minutes with 20ul of 250ug/ml human IgG (To block nonspecific binding), after which they were incubated 45 minutes on ice with 8 ul of I7662-15Y2 at 1ug/ml. 5. Cells were washed twice and incubated with S7973-88P Streptavidin (PE) after which they were washed three times, fixed and analyzed using a BD FACS Calibur. A net 40% sub population of the cells stained positive with a mean shift of 1.0 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml anti-IFN-gamma antibody. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
