- The Insulin Receptor (IR) is synthesized as a single polypeptide, which is subsequently cleaved to generate an extracellular alpha-chain and a transmembrane and intracellular beta-chain, tethered together by disulfide bonds. The beta-chain has multiple tyrosine phosphorylation sites, including three autophosphorylation sites at its activation loop. The overall structure of the IR is highly homologous to the IGF-I Receptor, except in their c-termini, where the two proteins diverge somewhat. The IR signals primarily by phosphorylating the Insulin Receptor Substrate (IRS) family of proteins, which creates docking sites for SH2-domain containing proteins. Insulin signaling is highly dependent on the PI3 Kinase pathway and Akt, which appear to mediate the functions of insulin. Applications: Suitable for use in ELISA and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Paraffin sections. Heat induced antigen retrieval with sodium citrate buffer, pH 6.0 is recommended. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
