- IKKa (IkB kinasea-, or IKK-1) is part of a large protein complex responsible for the inducible phosphorylation of IkB proteins. The same protein was originally identified as CHUK (conserved helix-loop-helix ubiquitous kinase), a serine/threonine kinase of unknown function. The human IKKa is a 85kD peptide that has been shown to activate NF-kB by phosphorylation of IkB proteins. IKKa interacts with its upstream kinase, NIK, and its downstream substrate, the IkB proteins. Mutations of IKKa in its kinase domain lead to a dominant-negative phenotype that suppresses TNFa and IL-1B induced NF-kB activaiton. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1:2000 Immunoprecipitation: 1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
