- Immunoglobuoin G is a molecule of about 150kD composed of four peptide chains. Each IgG contains two identical gamma heavy chains of about 51kD and two identical light chains of about 26kD, thus a tetrameric quaternary structure. The two heavy chains are linked to each other and to a light chain each by disulfide bonds. The resulting tetramer has two identical halves, which together form the Y-like shape. Each end of the fork contains an identical antigen binding site. The Fc regions of IgGs bear a highly conserved N-glycosylation site. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 1:20,000-1:500,000 has been assayed against 1ug of rat IgG in a standard capture ELISA using Streptavidin (HRP) and ABTS (A0030) as a substrate for 30 minutes at RT. Western Blot: 1:2000-1:10,000 Immunohistochemistry: 1:1000-1:1:5000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
