- 11β-Hydroxysteroid Dehydrogenase (Type 2) (11β-HSD2) catalyzes the conversion of cortisol (hydrocortisone) to the inactive glucocorticoid cortisone, thereby protecting the mineralcorticoid receptor from glucocorticoid excess. It is primarily expressed in the kidneys, but has also been detected in the colon, salivary glands, and fetal tissues, including the placenta, and is localized to the endoplasmic reticulum. Knockout of HSD11B2, the gene encoding 11β-HSD2, reduces fetal and placental growth as well as capillary development in mice. Loss-of-function mutations in HSD11B2 result in apparent mineralcorticoid excess (AME), an inborn error of metabolism characterized by hypertension, hypokalemia, and reduced plasma renin activity. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (FFPE): 1:100. Requires HIER using sodium citrate buffer pH6.0 prior to staining. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
