- Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs). Acetylation of lysine14 is commonly seen in genes that are being actively transcribed into RNA. Applications: Suitable for use in Western Blot, Dot Blot, Chromatin Immunoprecipitation and Flow Cytometry. Other applications not tested. Recommended Dilutions: Western Blot: 1:10,000. Detects acetylated histone H3 on 10ug of sodium butyrate treated HeLa cell lysates. Dot Blot: 1:1000 Chromatin Immunoprecipitation: Immunoprecipitated Histone H3, acetylated (Lys9) in 5ug of HeLa cell lysates Flow Cytometry: 0.25ug detected Histone H3, acetylated (Lys9) in 1x10e6 HEK293 cells Optimal dilutions to be determined by the researcher. Positive Control: HeLa cell lysates Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb I1904-46Q: IgG, H&L (HRP) Pab Gt x Rb Storage and Stability: May be stored at 4°C for short-term only.
