- Histone H3 is a core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Applications: Suitable for use in Dot Blot, Western Blot, Peptide Inhibition Assay. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-1:6000. Detects methylated Histone H3 (Lys9) in acid- extracted proteins from HeLa cells. Did not detect unmethylated recombinant Histone H3. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
