Extra Details:
Heparin Cofactor II (HCII) is a single chain glycoprotein produced in the liver and circulates in plasma at concentration of 50-100ug/ml (0.8-1.5uM). A member of the SERPIN family of proteinase inhibitors, HCII exhibits a relatively narrow specificity and is only known to inhibit the serine proteinases thrombin and chymotrypsin. The inhibitory activity toward thrombin is accelerated up to 1000-fold in the presence of optimal concentrations of heparin. Unlike Antithrombin, thrombin inhibition by HCII is also enhanced by dermatan sulphate. HCII has an apparent molecular weight of 65,600 daltons and interaction with thrombin results in a covalent 1:1 enzyme-inhibitor complex of 102,600 daltons (1-3). Applications: Suitable for use in ELISA and Immunoelectrophoresis. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Rz Ratio (Reinheitszahl, A403/A280): 0.46 Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
