- Extra Details:
- Heparin Cofactor II (HCII), also known as heparin cofactor A and dermatan sulfate cofactor, is a single chain glycoprotein produced in the liver and circulates in plasma at concentration of 50-100ug/ml (0.8-1.5uM). A member of the SERPIN family of proteinase inhibitors, HCII exhibits a relatively narrow specificity and is only known to inhibit the serine proteinases thrombin and chymotrypsin. The inhibitory activity toward thrombin is accelerated up to 1000-fold in the presence of optimal concentrations of heparin. Unlike Antithrombin, thrombin inhibition by HCII is also enhanced by dermatan sulfate. HCII has an apparent molecular weight of 65,600 daltons and interaction with thrombin results in a covalent 1:1 enzyme-inhibitor complex of 102,600 daltons (1-3). Applications: Suitable for use as capture antibody in ELISA for Heparin Cofactor II and its complexes. Also suitable for use in Immunodiffusion versus citrated plasma. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
