Extra Details:: Human growth hormone (hGH, somatotropin), a protein hormone from the anterior lobe of the pituitary gland, comprises a single polypeptide chain of 191 amino acid residues. The 20kD variant form of human growth hormone (20kD hGH) presents in extracts from pituitary glands and it differs from the major form of hGH (22kD, 191 amino acids) by the deletion of amino acid residues 32-46. The growth hormone amino acid sequence varies considerably between species, and non-primate growth hormones have little activity in man. Main functions of hGH and 20kD hGH are the stimulation of somatic and bone growth, as well as an increase in the size and mass of organs and tissues. Applications: Suitable for use in Neutralization, ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:250-1:500; Recombinant hGH and 20kD hGH were resolved by electrophoresis, transferred to PVDF membrane and probed with G9000-03 (1:500). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a DAB detection system. Neutralization: Measured by its ability to neutralize Growth hormone induced proliferation in Nb2-11 rat lymphoma cells. The neutralization ED50 is less than or equal to 2ug/ml in the presence of 2.5ng/ml recombinant protein human Growth hormone. Optimal dilutions to be determined by the researcher. Hybridoma: SP2/O myeloma cells with spleen cells from BALB/c mice. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
