- Human Granulocyte colony stimulating factor (GM-CSF) is a 14.6kD glycoprotein, which is produced by T cells, macrophages, fibroblasts and endothelial cells. GM-CSF is involved in the stimulation of proliferation and differentiation of granulocyte and macrophage progenitor cells. Applications: Suitable for use in Neutralization, Immunocytochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Immunocytochemistry: 8-25ug/ml in immersion fixed human peripheral blood mononuclear cells (PBMCs). Western Blot: 1ug/ml, non-reducing conditions only. Neutralization: Measured by its ability to neutralize GM‑CSF-induced proliferation in the TF‑1 human erythroleukemic cell line. Kitamura, T. et al. (1989) J. Cell Physiol. 140:323. The Neutralization Dose (ND50) is typically 0.075-0.45ug/ml in the presence of 0.1ng/ml recombinant Human GM‑CSF. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
