- GTP-binding proteins (G-proteins) play a critical role in signal transduction by coupling receptors to effectors. The intracellular consequences of G-protein activation include second messenger generation, protein phosphorylation, ion channel activation, gene induction, cell growth, and differentiation. Gi belongs to the pertussis toxin sensitive G-proteins. Gi consists of three different subtypes of alpha-subunits (alphai1, alphai2 and alphai3). Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Paraffin sections require HIER with citrate buffer, pH 6.0 Optimal dilutions to be determined by the researcher. Positive Control: Western Blot: L6 Cell Lysate Immunohistochemistry: Rat cerebellum Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
