- Filaggrins are an important class of the intermediate filament-associated proteins which interect with keratin intermediate filaments (Ifs) of terminally differentiating mammalian epidermis. Filaggrins exhibit wide species variations. Filaggrin also appears to be a target molecule for rheumatoid arthritis-specific auto-antibodies in humans. Filaggrin is expressed only in well differentiated keratinized epithelial cells. The synthesis of filaggrin varies in types of ichthyosis, and with viral, premalignant and malignant conditions. Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistology (FFPE): 2-4ug/ml for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Positive Control: Skin Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
