Extra Details:: Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested. Recommended Dilution: Immunocytochemistry: 10ug/ml shows positive immunostaining for Fak in Ref 52 cells fixed with 3.7% formaldehyde. Western Blot: 0.5-2ug/ml detects FAK in RIPA lysates from murine 3T3 and previously in human A431 RIPA cell lysate. 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with F0019-56 (1ug/ml). Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates FAK from 500ug of murine 3T3 RIPA lysate. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb I1904-46Q: IgG, H&L (HRP) Pab Gt xRb I1904-40D: IgG, H&L (FITC) Pab Gt xRb I1904-41: IgG, X-Adsorbed (FITC) Pab Gt xRb I1904-46N: IgG, H&L (FITC) Pab Gt xRb Positive Antigen Control: F4209 3T3 cell lysate; use 20mg per lane for minigels. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
