- Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested. Recommended Dilution: Immunocytochemistry: 10ug/ml shows positive immunostaining for Fak in Ref 52 cells fixed with 3.7% formaldehyde. Western Blot: 0.5-2ug/ml detects FAK in RIPA lysates from murine 3T3 and previously in human A431 RIPA cell lysate. 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with F0019-56 (1ug/ml). Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates FAK from 500ug of murine 3T3 RIPA lysate. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb I1904-46Q: IgG, H&L (HRP) Pab Gt xRb I1904-40D: IgG, H&L (FITC) Pab Gt xRb I1904-41: IgG, X-Adsorbed (FITC) Pab Gt xRb I1904-46N: IgG, H&L (FITC) Pab Gt xRb Positive Antigen Control: F4209 3T3 cell lysate; use 20mg per lane for minigels. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
