- erbB4/HER4 is the fourth member of the human epidermal growth factor receptor (EGFR) family that is frequently overexpressed in a variety of aggressive epithelial carcinomas. Its extracellular domain is most similar to the orphan receptor HER3, whereas its cytoplasmic kinase domain exhibits 79% and 77% identity with EGFR and HER2, respectively. HER4 is most predominantly expressed in several breast carcinoma cell lines, and in normal skeletal muscle, heart, pituitary, brain and cerebellum. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 detects erbB-4 on 10ug of A431 lysates. Immunohistochemistry: formalin-fixed, paraffin-embedded sections. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
