MaxLight™ 490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. The Eph receptors are the largest family of receptor tyrosine kinases. They can be grouped into two subclasses: the EphA and the EphB receptors. The ligands for the Eph receptors are the ephrins. The EphA receptors bind ephrin-A ligands, which are tethered to the membrane by a GPI linkage, and the EphB receptors bind ephrin-B ligands, which have a membrane spanning region. The Eph receptors are widely expressed during embryonic development. Ephrins and Eph receptors have been implicated in many developmental processes including neuronal network formation, guidance of cell migration, and axonal pathfinding. They remain expressed at lower levels in adult tissues and are upregulated in pathological conditions such as cancer. EphB4 is expressed in epithelial cells and in endothelial cells of embryonic veins, where its signaling mediates cell migration and proliferation via the PI3K pathway. It is upregulated in several types of tumors, including mammary tumors, colon cancer, and endometrial cancer. EphB4 has previously been designated Htk, Myk1, Tyro11 and Mdk2. Applications: Suitable for use in FLISA, Western Blot, Immunoprecipitation, Immunofluorescence, Immunocytochemistry and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Positive Controls: Western Blot: EphB4-transfected 293T cells and human MDA-MB-231 breast carcinoma cell lysates Immunohistochemistry: Human colon cancer tissues Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
