- Extra Details:
- This gene encodes the D1 subtype of the dopamine receptor. The D1 subtype is the most abundant dopamine receptor in the central nervous system. This G-protein coupled receptor stimulates adenylyl cyclase and activates cyclic AMP-dependent protein kinases. D1 receptors regulate neuronal growth and development, mediate some behavioral responses, and modulate dopamine receptor D2-mediated events. Alternate transcription initiation sites result in two transcript variants of this gene. The rat D1a receptor is a 446aa, G-protein coupled and transmembrane receptor protein. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications not tested. Recommended Dilutions: Western Blot (Chemiluminescence): 1-10ug/ml. Detects 2 bands of 50 and 75kD in solubilized hippocampal membranes. The 75kD band was not visible in striatum. ~100-400ug of protein/lane should be loaded to visualize the 75kD band that is present in relatively small amounts. ELISA: 0.5-1.0ug/ml using 1ug/ml of control peptide per well Immunohistochemistry: 2-10ug/ml on paraformaldehyde fixed tissue sections. D1R has been localized in various regions of the rat brain. Optimal dilution determined by the researcher. Positive Control: Rat striatal membranes (western blot) Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
