Extra Details:
DMP-1 (dentin matrix protein 1) is an acidic, 100-160kD member of the phosphophoryn family of extracellular matrix proteins. Mouse DMP-1 is found both intra- and extracellularly, where phosphorylated DMP-1 participates in bone mineralization. Secreted mature mouse DMP-1 is a 487 amino acid (aa) proteoglycan. It contains multiple serine, glutamic acid, and aspartic acid residues, a GAG attachment site (aa71-72) and an RGD motif (aa350-352). Multiple cleavage sites exist between aa205-253, creating multiple fragments ~37kD and 57kD in size. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.1ug/ml Immunohistochemistry: 10ug/ml detects DMP1 in E15.5 mouse embryos. Fix cells with PBS, 4% paraformaldehyde for 20 minutes at RT and block with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. Incubate cells with D8075-51 for 3 hours at RT followed by donkey anti-sheep lgG at RT for 1 hour. Between each step, wash cells with PBS, 0.1% BSA. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
