- Extra Details:
- During base excision repair (BER) DNA polymerase b (Pol b ) is recruited by APE1. Pol b has two distinct domains, the larger is the polymerase domain itself, whereas a small basic N-terminal domain contains an AP lyase activity that excises the abasic sugar-phosphate residue at the strand break. Pol b also interacts with XRCC1, DNA ligase III and poly(ADP-ribose) polymerase in the repair of DNA single strand breaks. Pol b plays a role in both BER pathways, "single-nucleotide" and "long patch". It is the most inaccurate of the six DNA polymerases found in mammalian cells. Overexpression of Pol beta, as found in some human tumors, could confer an increase in spontaneous mutagenesis. It has been further suggested that Pol b does not excise 5'-oxidized AP sites induced by reactive oxygen species (ROS). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:500 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
