- MaxLight™ 650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. The desmosome is subdivided into two regions. The plaque region lies adjacent to the plasma, and is believed to contain molecules that attach the intermediate filament cytoskeleton to the desmosome. The core region is composed of transmembrane glycoproteins that are thought to mediate cell-cell adhesion. Desmogleins and desmocollins are the main desmosomal transmembrane proteins. These desmosomal glycoproteins belong to the members of the cadherin family of adhesion molecules. Three different isoforms of both desmogleins and desmocollins have been identified, named as Desmoglein 1-3 and Desmocollin 1-3. Desmosomal cadherins showed differentiation-specific expression in the human epidermis, although the functional significance of this differential expression is not fully understood. Desmoglein 1 is synthesized exclusively in the suprabasal layers. The expression of Desmoglein 2 varies in the basal layers, depending on the certain types of epidermis. Desmoglein 3 is expressed in both the basal and the lower suprabasal compartments. Domain specific desmosomal and adherens junction-associated antibodies have been widely used in recent studies. Desmosomal cadherin antibodies are also useful for studying specific disease states and cell adhesion protein interactions. Applications: Suitable for use in Immunoprecipitation, Western Blot and Immunofluorescence. Other applications not tested. Recommended Dilutions: Immunoprecipitation: Native Optimal dilutions to be determined by the researcher. Positive Control: A431 cells Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
